However, compared with the recent progress of follicular stem cell research, detailed analysis of dermal papilla cells is virtually unknown. We have focused on a large chondroitin sulfate proteoglycan molecule, versican, and prove its pivotal role in hair follicle induction both in vitro and in vivo. First, we examined versican expression with hair cycling by in situ hybridization (mRNA) and immunohistochemistry (protein). Results clearly showed specific versican expression in anagen onset of dermal papilla cells implying a function role of versican in hair induction. We then generated transgenic lines with LacZ or green fluorescent protein reporter genes under a versican regulatory element (promoter) control, and confirmed dermal papilla-specific activation of the reporter. These transgenic lines were utilized to isolate the dermal papilla cell population by means of a fluorescent activated cell sorter. While freshly isolated dermal papilla cells were able to induce hair growth in a skin reconstitution assay when grafted with undifferentiated epidermal cells, this ability and versican expression were rapidly lost during passages in culture. In order to examine the functional role of versican in hair growth induction, versican full-length cDNAs were transfected into inactive passaged dermal papilla cells. Impressively, this forced expression of versican molecules partially restored hair inductivity of dermal papilla cells in a skin reconstitution assay. These results demonstrated the pivotal role of versican in hair induction both in vitro and in vivo and will connect the missing link between the signal from dermal papilla cells to follicular stem cells, and initiation of hair regeneration. We have also shown that the hair growth-promoting compound, cyclosporin A, can upregulate versican expression implying the application of a novel screening approach for hair growth-promoting compounds by monitoring versican expression as an indicator. Dus eigenlijk is DHT niet de ware boosdoener, maar het immuunsysteem. Deze valt de haarfollikels aan.

Wat zou er gebeuren als je cyclosporine voor 8 weken ofzo gebruikt?

Inderdaad, er staat iets over een product Folligen dat veelbelovend is/zal zijn. Nu begrijp ik ook het principe van de inflammation (ontstekingsreacties); AGA is een soort van auto-immuum proces. Het immuumsysteem valt de haarfollikels aan, omdat zij als niet-lichamelijk worden beschouwd door de aantasting van DHT. Auto-immuum gaat volgens mij vaak gepaard met ontstekingsreacties enzovoort. Vandoor ook dat men zich tegenwoordig vooral richt op de anti-inflammation en niet zozeer alleen meer op anti-androgeen.
En misschien is dit ook wel de reden waarom de rozemarijn shampoo zo goed bevalt. Rozemarijn schijnt ook (in bepaalde mate) de ontstekingsreacties te remmen. |

Oppassen met cyclosporine
Hee iedereen,
Pindakaas, ten eerste weer bedankt voor dit interessante artikel. Ik wilde alleen echt even waarschuwen voor ciclosporine / cyclosporine. Als je immuunsysteem platligt, kan je lichaam NERGENS tegen. Ik weet hoe makkelijk medicijnen tegenwoordig op internet te bestellen zijn, maar ga hier dus niet aan beginnen! Dat wilde ik even zeggen. |

AGA immuungevolg?
Nu ik me meer aan het verdiepen ben in cyclosporine, begin ik zelf meer en meer te twijfelen of de hele immuuntheorie (dat AGA wordt veroorzaakt door aanvallen van het immuunsysteem, ofwel inflammation) wel klopt. Uit de onderzoeken die ik heb gevonden lijkt het er meer op dat cyclo een algemeen haargroeieffect heeft, net zoals minoxidil. Het valt onder de zogenaamde "hypertrichotic" middelen, ofwel de middelen die een effect hebben op de haargroei. Uit de onderzoeken blijkt dat cyclo, net als minox, de anagene fase verlengt van iedere haar (ook lichaamsharen). Of dit komt door onderdrukking van het immuunsysteem is de vraag.
In een van de onderzoeken staat zelfs: These results suggest that [...] anagen hair induction caused by cyclosporin A and FK506 is not a result of immunosuppression.Dus wie weet werkt cyclo op soortgelijke wijze als minox en klopt de hele immuuntheorie niet. Er waren altijd een aantal bewijzen voor de immuntheorie:
Alleen bewijs 1) staat nog overeind, maar daar heb ik me nog nooit in verdiept. Ik zal eens kijken of daar wat over te vinden is. |
